RESUMO

OBJECTIVES: To test the feasibility, acceptability, and potential efficacy of a mHealth intervention tailored for Chinese immigrant families with type 2 diabetes (T2D). METHODS: We conducted a pilot randomized controlled trial (RCT) with baseline, 3-, and 6-month measurements. Participating dyads, T2D patients and families/friends from NYC, were randomized into the intervention group (n = 11) or the wait-list control group (n = 12). Intervention includes 24 videos covering T2D self-management, behavioral techniques, and family-oriented sessions. Feasibility and acceptability were measured respectively by the retention rate and video watch rate, and a satisfaction survey. Patients' HbA1c, weight, and self-management were also assessed to test potential efficacy. RESULTS: Most T2D patients (n = 23; mean age 56.2±9.4 years; 52.2% male) and families/friends (n = 23, mean age 54.6±11.2 years; 52.2% female) had high school education or less (69.6% and 69.6%), annual household income < $25,000 (65.2% and 52.2%), and limited English proficiency (95.7% and 95.7%). The retention rates were not significantly different between the intervention and the control groups for both the patients (90.91% vs 83.3%, p = 0.589); and their families/friends (3-month: 90.9% vs 75%, p = 0.313; 6-month: 90.9% vs 83.3%, p = 0.589). The mean video watch rate was 76.8% (7%). T2D patients and families/friends rated satisfaction as 9.4 and 10 out of 10, respectively. Despite no between-group differences, the intervention group had significantly lower HbA1c (p = 0.014) and better self-management (p = 0.009), and lost 12 lbs. on average at 6 months (p = 0.079), compared to their baseline levels. CONCLUSIONS: A culturally-tailored, family-based mHealth intervention is feasible and acceptable among low-income, limited English-proficient Chinese families with T2D in NYC. Significant changes in HbA1c and self-management within the intervention group indicate this intervention may have potential efficacy. Given the small sample size of this study, a future RCT with adequate power is needed to test efficacy.

Theory and experiment have revealed that spin-orbit coupling (SOC) strongly depends on the relativistic effect in topological insulators (TIs), while the influence of orbitals is always ignored. Herein, we provide a direct way of controlling effective SOC with the help of orbital effects, reducing the dependence on elements. Taking 5d W2CO2 and 4d Mo2CO2 MXenes as a specific example, we predict that by decreasing the hybridization strength of W atoms with C or O atoms in 2D W2CO2, the nontrivial bandgaps at the Γ-point are directly enhanced. The weak hybridization of W atoms with ligand elements enhances the electron localization of degenerate d-orbitals of three groups under the triangular prism crystal field, inducing stronger on-site Coulomb repulsion that enhances orbital polarization as well as boosts the SOC effect. Meanwhile, similar results have also been observed in 4d Mo2CO2. This implies that the orbital effects are an efficient and straightforward way to control the nontrivial bandgap in 2D MXene TIs. Our work not only provides an alternative perspective on designing large nontrivial bandgaps but also brings a possibility to control the SOC effect for TI devices.

The recent surge in plant genomic and transcriptomic data has laid a foundation for reconstructing evolutionary scenarios and inferring potential functions of key genes related to plants' development and stress responses. The classical scheme for identifying homologous genes is sequence similarity-based searching, under the crucial assumption that homologous sequences are more similar to each other than they are to any other non-homologous sequences. Advances in plant phylogenomics and computational algorithms have enabled us to systemically identify homologs/orthologs and reconstruct their evolutionary histories among distantly related lineages. Here, we present a comprehensive pipeline for homologous sequences identification, phylogenetic relationship inference, and potential functional profiling of genes in plants. Key features • Identification of orthologs using large-scale genomic and transcriptomic data. • This protocol is generalized for analyzing the evolution of plant genes.

The development of cost-effective, biocompatible soft wound dressings is highly desirable; however, conventional dressings are only designed for flat wounds, which creates difficulty with promising healing efficiency in complex practical conditions. Herein, we developed a tough, adhesive biomimetic hyaluronic acid methacryloyl hydrogels composed of chemically crosslinked hyaluronic acid methacryloyl (HAMA) network and poly(N-hydroxyethyl acrylamide) (PHEAA) network rich in multiple hydrogen bonding. Due to the multiple chemical crosslinking sites (acrylamide groups) of HAMA; the bulk HEMA/PHEAA hydrogels presented significant enhancements in mechanical properties (∼0.45 MPa) than common hyaluronic acid hydrogels (<0.1 MPa). The abundant hydrogen bonding also endowed the resultant hydrogels with extremely high adhesiveness on many nonporous substrates, including glass and biological tissues (e.g., heart, liver, lung, kidney, stomach, and muscle), with a considerable interfacial toughness of ∼1432 J m-2. Accordingly, since both natural hyaluronic acid derivative polymers and hydrophilic PHEAA networks are highly biocompatible, the hydrogel matrix possesses good blood compatibility (<5% of hemolysis ratio) and satisfies the general dressing requirements (>99% of cell viability). Based on these physicochemical features, we have demonstrated that this adhesive hydrogel, administered in the form of a designed patch, could be applied to wound tissue healing by promoting epithelialization, angiogenesis, and collagen deposition. We believe that our proposed biomimetic hydrogel design holds great potential for wound repair and our developed HAMA/PHEAA hydrogels are extremely promising for the next-generation tissue healings in emergency situations.

OBJECTIVE: Analysis of the risk factors affecting hip function and complications after femoral neck system (FNS) surgery for femoral neck fractures is of great significance for improving the procedure's efficacy. METHODS: The data of patients with femoral neck fractures who underwent FNS surgery in our hospital between October 2019 and October 2020 were retrospectively analyzed. Age, gender, time from injury to operation, fracture classification, operation time, fracture reduction, and postoperative weight-bearing time information were set as potential factors that may affect the results. Hip Harris scores were performed at 12 months postoperatively, and postoperative complication data (e.g., femoral head necrosis, nonunion, and femoral neck shortness) were collected. The risk factors affecting hip function and complications after FNS surgery were predicted using linear and logistic regression analyses. RESULTS: A total of 69 cases of femoral neck fracture were included, with an average age of 56.09 ± 11.50 years. The linear analysis demonstrated that the age and fracture type of the patients were the risk factors affecting the Harris score of the hip joint after FNS surgery. Older patients with displaced femoral neck fractures had an inferior postoperative hip function. In addition, fracture type, reduction of the femoral neck, and postoperative weight-bearing significantly impacted postoperative complications. Displaced fractures, negative fixation, and premature weight-bearing (< 6 weeks) were risk factors for postoperative complications. The Harris score of patients with a shortened femoral neck in the included cases was not significantly different from that of patients without shortening (P = 0.25). CONCLUSIONS: Advanced age and fracture type are important evaluation indicators of the Harris score after FNS internal fixation of femoral neck fractures in young patients. Fracture type, fracture reduction, and postoperative weight-bearing time are risk factors for complications after FNS.

The outbreak of the COVID-19 pandemic has brought massive shifts in human activities through a global blockade, directly affecting wildlife survival. However, the indirect impacts of changes in human activities are often easily overlooked. We conducted surveys of Reeves's Pheasant (Syrmaticus reevesii) and its sympatric species by camera traps in forest-type nature reserves in three different scenarios: pre-lockdown, lockdown and post-lockdown. An increase in livestock activities observed during the lockdown and post-lockdown period in our study area provided us an opportunity to investigate the indirect impact of the lockdown on wildlife. The pre-lockdown period was used as a baseline to compare any changes in trends of relative abundance index, activity patterns and temporal spacing of targeted species and livestock. During the lockdown period, the relative abundance index of livestock increased by 50% and there was an increase in daytime activity. Reeves's Pheasant showed avoidance responses to almost all sympatric species and livestock in three different periods, and the livestock avoidance level of Reeves's Pheasant during the lockdown period was significantly and positively correlated with the relative abundance index of livestock. Species-specific changes in activity patterns of study species were observed, with reduced daytime activities of Hog Badger and Raccoon Dog during and after the confinement periods. This study highlights the effect of the COVID-19 lockdown on the responses of wildlife by considering the changes in their temporal and spatial use before, during and after lockdown. The knowledge gained on wildlife during reduced human mobility because of the pandemic aids in understanding the effect of human disturbances and developing future conservation strategies in the shared space, to manage both wildlife and livestock.

Metformin can enhance cancer cell chemosensitivity to anticancer drugs. IGF-1R is involved in cancer chemoresistance. The current study aimed to elucidate the role of metformin in osteosarcoma (OS) cell chemosensitivity modulation and identify its underlying mechanism in IGF-1R/miR-610/FEN1 signalling. IGF-1R, miR-610, and FEN1 were aberrantly expressed in OS and participated in apoptosis modulation; this effect was abated by metformin treatment. Luciferase reporter assays confirmed that FEN1 is a direct target of miR-610. Moreover, metformin treatment decreased IGF-1R and FEN1 but elevated miR-610 expression. Metformin sensitised OS cells to cytotoxic agents, while FEN1 overexpression partly compromised metformin's sensitising effects. Furthermore, metformin was observed to enhance adriamycin's effects in a murine xenograft model. Metformin enhanced OS cell sensitivity to cytotoxic agents via the IGF-1R/miR-610/FEN1 signalling axis, highlighting its potential as an adjuvant during chemotherapy.

Apical hooks are functional innovations only observed in angiosperms, which effectively protect the apical meristems out of damage during plant seedlings penetrating soil covers. Acetyltransferase like protein HOOKLESS1 (HLS1) in Arabidopsis thaliana is required for hook formation. However, the origin and evolution of HLS1 in plants are still not solved. Here, we traced the evolution of HLS1 and found that HLS1 originated in embryophytes. Moreover, we found that Arabidopsis HLS1 delayed plant flowering time, in addition to their well-known functions in apical hook development and newly reported roles in thermomorphogenesis. We further revealed that HLS1 interacted with transcription factor CO and repressed the expression of FT to delay flowering. Lastly, we compared the functional divergence of HLS1 among eudicot (A. thaliana), bryophytes (Physcomitrium patens and Marchantia polymorpha) and lycophyte (Selaginella moellendorffii). Although HLS1 from these bryophytes and lycophyte partially rescued the thermomorphogenesis defects in hls1-1 mutants, the apical hook defects and early flowering phenotypes could not be reversed by either P. patens, M. polymorpha or S. moellendorffii orthologs. These results illustrate that HLS1 proteins from bryophytes or lycophyte are able to modulate thermomorphogenesis phenotypes in A. thaliana likely through a conserved gene regulatory network. Our findings shed new light on the understanding of the functional diversity and origin of HLS1, which controls the most attractive innovations in angiosperms.

Objective: To investigate the correlation analysis of larger side bone cement volume/vertebral body volume ratio (LSBCV/VBV%) with adjacent vertebral compression fracture (AVCF) in percutaneous vertebroplasty (PVP) for osteoporotic vertebral compression fracture (OVCF). Methods: A retrospective analysis of 245 OVCF patients who underwent PVP treatment from February 2017 to February 2021, including 85 males and 160 females. The age ranged from 60 to 92 years, with a mean of (70.72 ± 7.03) years. According to whether AVCF occurred after surgery, they were divided into 38 cases in the AVCF group (fracture group) and 207 cases in the no AVCF group (non-fracture group). The correlation between gender, age, bone mineral density (BMD), body mass index (BMI), thoracolumbar segment fracture, bone cement disc leakage, LSBCV, bone cement volume (BCV), VBV, LSBCV/VBV ratio (LSBCV/VBV%), and BCV/VBV% and AVCF were analyzed in both groups. Risk factors for AVCF after PVP were analyzed by multifactorial logistic regression, and then the receiver operating characteristic curves (ROC curves) were plotted to identify the critical value of LSBCV/VBV%. Results: 38 patients (15.5%) developed AVCF postoperatively. Univariate analysis showed that BMD, bone cement disc leakage, LSBCV, and LSBCV/VBV% were risk factors for AVCF after PVP (P<0.05), while gender, age, BMI, thoracolumbar segment fracture, BCV, VBV, and BCV/VBV% were not significantly different in both groups (P>0.05). Multifactorial logistic regression analysis revealed that BMD, bone cement disc leakage, and LSBCV/VBV% were independent risk factors for AVCF after PVP (P<0.05). According to the ROC curve, the LSBCV/VBV% had an area under the curve of 71.6%, a sensitivity and specificity of 89.5% and 51.7%, respectively, and a critical value of 13.82%. Conclusion: BMD, bone cement disc leakage and LSBCV/VBV% are independent risk factors for AVCF after PVP. With LSBCV/VBV at 13.82%, the incidence of AVCF significantly increased.

The excessive migration of small molecular plasticizers in solid propellants may lead to debonding and changes in combustion characteristics, affecting the safety of solid rocket motors. Herein, two functionalized graphene oxides (GO) were used to enhance the anti-migration performance of EPDM insulation. GO, 3-Aminopropyltriethoxysilane-modified GO (AGO) and octadecylamine-modified GO (HGO) were filled into EPDM to fabricate EPDM insulation. The anti-migration properties and migration kinetics of EPDM insulations were studied using immersion tests. Moreover, the mechanical properties, including the tensile properties, crosslink density, hardness, and aging resistance of different EPDM insulations, were also explored. Compared with GO, AGO, and HGO obviously enhanced the anti-migration and mechanical properties of the EPDM insulations. This study shows that the anti-migration performance of EPDM insulation can be enhanced by functionalized GO.

A two-dimensional MXene (Ta4C3) was innovatively used herein to modulate the space group and electronic properties of vanadium oxides, and the MXene/metal-organic framework (MOF) derivative VO2(B)@Ta4C3 with 3D network cross-linking was prepared, which was then employed as a cathode to improve the performance of aqueous zinc ion batteries (ZIBs). A novel method combining HCl/LiF and hydrothermal treatments was used to etch Ta4AlC3 to obtain a large amount of accordion-like Ta4C3, and the V-MOF was then hydrothermally grown on the surface of the stripped Ta4C3 MXene. During the annealing process of V-MOF@Ta4C3, the addition of Ta4C3 MXene liberates the V-MOF from agglomerative stacking, allowing it to show additional active sites. More significantly, Ta4C3 prevents the V-MOF in the composite structure from converting into V2O5 of space group Pmmn but into VO2(B) of space group C2/m after annealing. A considerable advantage of VO2(B) for Zn2+ intercalation is provided by the negligible structural transformation during the intercalation process and the special tunnel transport channels, which have an enormous area (0.82 nm2 along the b axis). According to first-principles calculations, there is a strong interfacial interaction between VO2(B) and Ta4C3, which deliver remarkable electrochemical activity and kinetic performances for the storage of Zn2+. Therefore, the ZIBs prepared with the VO2(B)@Ta4C3 cathode material exhibit an ultra-high capacity of 437 mA h·g-1 at 0.1 A·g-1 while showing good cycle performance and dynamic performance. This study will offer a fresh approach and a reference for creating metal oxide/MXene composite structures.

OBJECTIVE: The purpose of this study was to compare the biomechanical and clinical results of two surgical methods for the treatment of vertical femoral neck fractures: Femoral neck system (FNS) and traditional three cannulated cancellous screws (CCS). METHODS: First, we developed three different vertical femoral neck fracture models for the finite element analysis, with angles of 55°, 65°, and 75°, respectively. Two experimental groups were set up: the FNS group and the CCS group. Each fracture group was tested under axial loads of 2100 N to measure the femur's displacement, Von Mises stress (VMS), and its internal fixation components. Secondly, we retrospectively included the cases of vertical femoral neck fractures with FNS and CCS in our hospital from May 2019 to May 2021. In this study, we compared the duration of intraoperative fluoroscopy, operative time, hospital stay, fracture healing time, Hemoglobin loss, Harris score of hip joint function, and postoperative complications among patients undergoing hip joint replacement. RESULTS: In terms of finite element analysis, FNS has better anti-displacement stability than CCS at 55°and 65°, while FNS is greater than CCS in Von Mises stress. Clinically, we followed up on 87 patients for an average of 12 months. FNS was superior to traditional CCS in fracture healing time, operation time, fluoroscopy duration, fracture healing time, and Harris hip function score. CONCLUSION: FNS is superior to traditional CCS in biomechanical and clinical aspects of treating vertical femoral neck fractures. There is potential for FNS to become a new treatment option for vertical femoral neck fractures.

Auxin inactivation is critical for plant growth and development. To develop plant growth regulators functioning in auxin inactivation pathway, we performed a phenotype-based chemical screen in Arabidopsis and identified a chemical, nalacin, that partially mimicked the effects of auxin. Genetic, pharmacological, and biochemical approaches demonstrated that nalacin exerts its auxin-like activities by inhibiting indole-3-acetic acid (IAA) conjugation that is mediated by Gretchen Hagen 3 (GH3) acyl acid amido synthetases. The crystal structure of Arabidopsis GH3.6 in complex with D4 (a derivative of nalacin) together with docking simulation analysis revealed the molecular basis of the inhibition of group II GH3 by nalacin. Sequence alignment analysis indicated broad bioactivities of nalacin and D4 as inhibitors of GH3s in vascular plants, which were confirmed, at least, in tomato and rice. In summary, our work identifies nalacin as a potent inhibitor of IAA conjugation mediated by group II GH3 that plays versatile roles in hormone-regulated plant development and has potential applications in both basic research and agriculture.

MAIN CONCLUSION: We reveal that transcription factors TOE1 and TOE2 directly inhibit the transcription of EIN3. Ethylene triggers leaf abscission and senescence during plant aging. Previous studies have shown that the transcription of ETHYLENE INSENSITIVE 3 (EIN3), which encodes a key transcription factor in ethylene signaling, is gradually upregulated during plant aging. However, it is still unknown how plants transmit their age information to achieve transcriptional control of EIN3. Here, we report that the EAR-like motif-containing transcription factors TARGET OF EAT 1 (TOE1) and its homolog TOE2 directly associated with the EIN3 promoter. The transcription of EIN3 is further enhanced in mutants of toe1 toe2 during plant aging. TOE1/TOE2 are tightly controlled by canonical microRNA 172 (miR172)-mediated plant aging signaling, which result in a decline in TOE1/TOE2 expression during aging. These results illustrate that during plant aging, the reduced expressions of TOE1/TOE2 trigger an upregulation of EIN3. Next, we took advantage of EIN3-regulated de novo root regeneration (DNRR) as an age-controlled phenotype to dissect the biological function of this regulatory circuit. The DNRR rates in toe1 toe2 are more severely decreased with plant aging; however, the simultaneous loss of ein3 and eil1 (toe1 toe2 ein3 eil1 quadruple mutants) almost completely rescued the DNRR defects. Taken together, our findings show that the plant age-regulated TOE transcription factors precisely integrate plant age information and developmental programs through direct protein-DNA interactions.

Unlike animals, plant development is plastic and sensitive to environmental changes. For example, Arabidopsis thaliana seedlings display distinct growth patterns when they are grown under different light or temperature conditions. Moreover, endogenous plant hormone such as ethylene also impacts seedling morphology. Ethylene induces hypocotyl elongation in light-grown seedlings but strongly inhibits hypocotyl elongation in etiolated (dark-grown) seedlings. Another characteristic ethylene response in etiolated seedlings is the formation of exaggerated apical hooks. Although it is well known that high ambient temperature promotes hypocotyl elongation in light-grown seedlings (thermomorphogenesis), ethylene suppresses thermomorphogenesis. On another side, high ambient temperature also inhibits the ethylene-responsive hypocotyl shortening and exaggerated hook formation in etiolated seedlings. Therefore, the simplest phytohormone ethylene exhibits almost the most complicated responses, depending on temperature and/or light conditions. In this review, we will focus on two topics related to the main theme of this special issue (response to high temperature): (1) how does high temperature suppress ethylene-induced seedling morphology in dark-grown seedlings, and (2) how does ethylene inhibit high temperature-induced seedling growth in light-grown seedlings. Controlling ethylene biosynthesis through antisense technology was the hallmark event in plant genetic engineering in 1990, we assume that manipulations on plant ethylene signaling in agricultural plants may pave the way for coping with climate change in future.

Antimony sulfide (Sb2S3), an emerging material for photovoltaic devices, has drawn growing research interest due to its inexpensive and high-throughput device production. In this study, the material and defect properties of Sb2S3 thin films prepared by the vapor transport deposition (VTD) method at different working pressures were studied. Solar cells based on a structure of glass/ITO/CdS/Sb2S3/Au were fabricated. The working pressure showed a significant effect on the device's performance. The current density versus voltage measurement and scanning electron microscopy analysis outcome were utilized to investigate the photovoltaic and microstructural properties in the samples. The compositional analysis by energy dispersive X-ray spectroscopy measurement confirmed the Sb/S ratio as 2:2.8 for the thin films. The identification and characterization of the defects present in Sb2S3 thin films were performed via admittance measurements. Compared to the defect density, the defect energy level was found to inherit a more important role in the device's performance. The best solar cell performance with better crystal quality, lower defect density, and longer capture lifetime was achieved under the substrate working pressure of 2 Pa. The highest efficiency was found to be 0.86% with Voc=0.55V, Jsc=5.07mA/cm2.

Kinases are indispensable signaling components. Radioactive-based phosphorylation assays are widely used but require specific protective equipment and safety trainings. Here, we present a Phos-tag-based non-radioactive kinase assay to study Arabidopsis kinase activities. We expressed and purified both kinase and substrate proteins from E. coli cells and then used the Phos-tag technology to detect the kinase activities under either different temperatures or chemical treatments. This non-radioactive approach is environmentally friendly and applicable to other kinases and organisms. For complete details on the use and execution of this protocol, please refer to Lin et al. Objective: The study aimed to investigate the effect of the type of bone cement distribution on clinical outcomes following percutaneous vertebroplasty (PVP) for osteoporotic vertebral compression fractures (OVCF) in the elderly. Methods: Retrospective analysis of 160 patients diagnosed with OVCF who underwent PVP treatment from March 2018 to December 2020. Based on the kind of postoperative bone cement distribution, bone cement was classified as types I, II, III, IV, and V. Visual Analog Scale (VAS), Oswestry Disability Index (ODI), Cobb angle, anterior vertebral height ratio, refracture rate of injured vertebrae, and incidence of adjacent vertebral fractures were compared for the five types before and after three days, and one year of operation. Results: VAS and ODI at three days and one year postoperative were significantly lower than those preoperative (P < 0.05) for all five distribution types. VAS and ODI for types I, II, and III were lower at one year postoperatively than for types IV and V (P < 0.05). There was no significant difference in Cobb angle and anterior vertebral body height ratio between preoperative and three days postoperative groups (P < 0.05); however, there were significant differences between three days and one-year postoperative and preoperative groups (P < 0.05). Following one year of surgery, the Cobb angle and the anterior vertebral height ratio of types IV and V were significantly different from those of types I, II, and III (P < 0.05), and there was a statistically significant difference between types IV and V (P < 0.05). In terms of the incidence of injured vertebral refractures and adjacent vertebral fractures, the evenly distributed types I, II, and III were significantly lower than the unevenly distributed types IV and V, and the incidence of type V was higher (P < 0.05). Conclusions: The clinical efficacy of cement distribution following PVP of types I, II, and III is better than that of types IV and V, which can better relieve pain with long-lasting efficacy and minimize the occurrence of refractures of injured vertebrae and adjacent vertebral body fractures.
